Discovery of novel antifungal compounds produced by Streptomyces spp
This project aims to discover novel antifungal compounds from Streptomyces spp. held in a
locally maintained strain library. The specific objectives are:
1. Resuscitate a collection of 100 Streptomyces strains stored at -80°C in glycerol stocks,
confirming viability on ISP2 agar, SFM agar , NL-5 plates
2. Screen these resuscitated strains for antifungal activity against “Saccharomyces cerevisiae” VL6-48 using agar overlay assays.
3. Characterize bioactive strains by:
(i) conducting bioassays against Candida albicans CA6 to assess clinical relevance
(ii) amplifying and sequencing 16S rRNA and rpoB genes via PCR for phylogenetic identification
(iii) sequencing genomes of 4 top isolates using Oxford Nanopore MinION
(iv) bioinformatically analysing secondary metabolite gene clusters with antiSMASH 7.0 to identify novel antifungal biosynthesis
pathways. ...
